Selank, GABA and anxiety experiments

The molecular experiments and the rat behaviour experiment make an awkward pair. I want the membrane and behavioural results explained together.

Rat-brain membrane binding

GABA helps quiet nerve activity. A 2018 review of Selank’s molecular activity discusses rat-brain membrane experiments in which Selank altered GABA binding. Effects depended on the receptor subtype and the concentration, and Selank could block the modulatory effects of diazepam and olanzapine in those assays.

That result initially suggested a simple opposition to me: one substance obstructing the influence of another. The whole-animal experiment complicates it. The binding assay measures what happens at membrane binding sites under particular conditions, leaving an animal's behaviour outside its frame.

Stressed rats given Selank with diazepam

In the 2017 unpredictable chronic mild stress experiment in rats, Selank together with diazepam reduced anxiety-like behaviour more effectively than either alone, bringing behaviour toward its pre-stress level.

I cannot forecast the behavioural result from that membrane finding. Different preparations, concentrations and outcomes leave plenty to explain. It is an interesting interaction to investigate, with human combination safety still untested. A better rat behavioural result provides no safe recipe for mixing medicines.

Calling Selank a gentler diazepam sounds premature to me. The review describes distinct, overlapping actions; the behavioural experiment tests a particular pairing in stressed animals. Neither gives a complete human pharmacological profile.

Gene-expression experiments

A 2016 rat frontal-cortex experiment found changes in expression of 45 genes after one hour and 22 after three hours. The changes correlated positively with those produced by GABA itself. These measurements concern copied genetic instructions connected with nerve signalling.

The 2008 rat hippocampus study also reported increased BDNF expression after intranasal Selank. BDNF is involved in nerve growth and maintenance. To me, the BDNF change supplies a molecular lead; a statement about improved human memory needs a different outcome measure.

Patient immune assays and tuftsin

Selank is a stabilised synthetic relative of tuftsin, a short fragment of an antibody protein. Added amino acids help it resist breakdown. That immune-system origin makes its 2008 patient immune findings less of a detour than they first appear.

The patients had anxiety-asthenic disorders, involving anxiety with weakness and exhaustion. The study found changes in the Th1/Th2 balance of immune signals and in IL-6 expression. Those are immune measurements in a specific patient group. They leave the overall health meaning of the immune shift and its long-term consequences unresolved.

Selank’s biological reach intrigues me. An account limited to feeling calm leaves out a whole set of measurements the human study actually made.

Selank accounts and risks

People online describe calmer feelings, easier social situations and steadier focus. Other accounts mention no clear change, drowsiness, headache or nasal irritation. These anecdotes are not clinical evidence, and their favourable claims about dependence or lasting safety have no long human follow-up behind them.

Selank has registration as an anxiety medicine in Russia and no FDA or EMA approval. Outside that registered medical setting, research material comes without the medicine assurances for identity, contents and sterility. Long-term safety and interactions involving sedatives, opioid-active drugs and medicines affecting serotonin or dopamine remain unresolved.

Pregnancy, breastfeeding and significant existing illnesses lack safety studies. Persistent anxiety calls for assessment by a qualified clinician. A substantial human follow-up would do more to reassure me than another assurance of “calm without sedation.”
